Cloning and Nucleotide Sequence Analysis of a 22 - kDa Kunitz - type Potato Proteinase Inhibitor Genomic DNA

Abstract
A genomic DNA, designated POTPIP340A, of the 22-kDa Kunitz-type potato proteinase inhibitor (22kDa KPPI) gene was identified, screened, and characterized. The POTPIP340A showed 91.3% homology with the 22-kDa cDNA, p34021, and more than 90% homology to other cDNA, PKP1. Whereas, there was no significant DNA sequence homology with proteinase inhibitors including proteinase inhibitor I and II genes. A transcription initiation site, 5¢¥ UTR, and promoter sequence have not been identified in the genomic DNA clone based on the results of DNA homology analysis by using the BestFit program in GCG package. Therefore, this cloned genomic DNA, POTPIP340A, was possibly a pseudogene of the 22-kDa KPPI gene.
